For river fishing on the James I use about 60 degrees, the current and heavy sinkers pulls em down to about 45.  While bank fishing for  flatheads using J hooks or Kahles, I set the poles from zero to 30 degrees depending on the softness of the tip.  If I am using circles then the angle is from 30 to 60 degrees, again depending on the softness of the tip.

On my boat, I use saltwater down easters, its the only rod holder built for fish up to 85 lbs and its big advantage is being able to release the rod into your hands under heavy pressure.  If you are going to build the ultimate rod holder then look at the functionality of the down easter, then build the 360 swiveling and 90 degree elvation into it.
